摘要
目的研究急性染锰对大鼠基底前脑斜角带垂直臂核(VDB)、斜角带水平臂核(HDB)区乙酰胆碱转移酶(ChAT)表达的影响及牛磺酸的干预作用。方法无特定病原体(SPF)级雄性SD大鼠随机分为:对照组,染锰组,预防组,治疗组,治疗对照组。ChAT免疫组化染色观察基底前脑VDB、HDB区乙酰胆碱能神经细胞形态与数量变化。结果 VDB区:染锰组ChAT阳性细胞数低于对照组,差异有统计学意义(P 0.05);预防组ChAT阳性细胞数与对照组相比,差异无统计学意义(P 0.05);预防组、治疗对照组ChAT阳性细胞数均高于染锰组,差异均有统计学意义(P 0.05);治疗组ChAT阳性细胞数高于治疗对照组,差异有统计学意义(P 0.05)。HDB区:染锰组、预防组ChAT阳性细胞数均低于对照组,差异均有统计学意义(P 0.05);预防组、治疗对照组ChAT阳性细胞数均高于染锰组,差异均有统计学意义(P 0.05);治疗组ChAT阳性细胞数与治疗对照组相比,差异无统计学意义(P 0.05)。结论大鼠急性染锰后,VDB、HDB区ChAT表达显著减少,牛磺酸预防能有效提高VDB、HDB区ChAT表达,牛磺酸治疗能有效提高VDB区ChAT表达。治疗对照组VDB、HDB区ChAT表达有所提高,说明短期染锰停止以后,大鼠基底前脑可能会逐渐修复损伤。
Objective To study the effect of acute Mn-treatment on the expression of acetyloholine transferase (CHAT) in nucleus of the vertical limb of the diagonal band (VDB) and nucleus of the horizontal limb of the diagonal hand(HDB) of rat and interference by taurine. Method SPF SD rats were randomly assigned into 5 groups: control, Mn-treated, taurine-prevented, taurine-treated, taurine-treated control. We stained the specimens by means of immunohistochemistry and then observed them through microscope and recorded counts of ChAT positive neurons in VDB and HDB of rats. We also took pictures for these neurons. Results In VDB: The count of ChAT positive neurons of Mn-treated group was markedly lower than those of control (P〈0.05). and not significantly different with the taurine-prevented group (P〉0.05). The counts of ChAT positive neurons of taurine-prevented and taurine-treated control groups were markedly higher than those of Mn-treated group(P〈0.05). The count of ChAT positive neurons of taurine-treated was markedly higher than that of taurine-treated control (P〈 0.05). In HDB: The counts of ChAT positive neurons of Mn-treated and taurine-prevented group were markedly lower than those of control (P〈 0.05). The counts of ChAT positive neurons of taurine-prevented and taurine-treated control groups were markedly higher than those of Mn-treated group (P〈0.0). The counts of ChAT positive neurons of taurine-treated and taurine-treated control group had no significant difference (P〉0.05). Conclusion MnC12.4H20 could acutely reduce the expression of ChAT in VDB and HDB of rats. Taurine could improve this impaired expression in prevented group, but could only improve the impaired expression of ChAT in VDB in treated group. The counts of ChAT positive neurons of VDB and HDB in taurine-treated control were also improved. So the impairment of ChAT in VDB and HDB of rat after acute Mn-treatment for short time may recover by themselves gradually.
