摘要
目的观察在不同浓度脂多糖(LPS)干预下,热休克蛋白(HSP)27在人牙周膜成纤维细胞(HPLFs)损伤过程中的表达情况。方法取原代培养至3~8代的HPLFs,以LPS梯度浓度干预,采用免疫组织化学方法检测HSP27在HPLFs中的表达。结果 HSP27在正常HPLFs中呈阴性表达,LPS干预后的HPLFs中HSP27呈胞质阳性表达,且随干预浓度增高表达量有逐渐增高的趋势。结论 HSP27在LPS介导的HPLFs炎症损伤中发挥重要的作用,其发生机制有待进一步研究。
Objective To investigate the expression of heat shock protein(HSP)27 in injury of human periodontal ligament fibroblasts (HPLFs) induced by different concentrations of lipopolysaccharide (LPS). Methods The 3th-8th passages of cultured HPLFs were treated with serially diluted LPS. Immunohistochemical assay was used to determine the expression of HSP27 in HPLFs. Results Expression of HSP27 was negative in normal HPLFs, and was positive in the cytoplasma of LPS-intervened HPLFs in a concentration-dependent manner. Conclusion HSP27 plays an important role during LPS-induced inflammation in HPLFs, although the underlying mechanisms remain to be further investigated.
