摘要
目的检测Kiss-1基因在5种不同转移习性的人鼻咽癌细胞株CNE-1、CNE-2Z、SUNE-1、SUNE-1-5-8F、SUNE-1-6-10B中的蛋白和mRNA水平的表达情况。方法应用逆转录酶聚合酶链式反应(RT-PCR)和免疫组织化学法在mRNA和蛋白水平检测5种细胞株中Kiss-1基因的表达。结果 5种细胞株中Kiss-1蛋白的表达皆为阳性,在低转移性细胞株SUNE-1-6-10B中Kiss-1基因在蛋白水平的表达显著高于其他具有较高转移习性细胞株中的表达(P<0.01)。5种细胞株中均扩增出Kiss-1 mRNA,在低转移性细胞株SUNE-1-6-10B中Kiss-1基因在mRNA水平的表达显著高于其他具有较高转移习性细胞株中的表达(P<0.01)。结论 Kiss-1基因在低转移性人鼻咽癌细胞株细胞中高表达,而在高转移性人鼻咽癌细胞株细胞中低表达,提示了该基因可能在鼻咽癌肿瘤细胞转移机制中发挥重要的抑制作用。
Objective To assessed the expression of Kiss-1 genes in human nasopharyngeal cell lines with the different metastasis ability(CNE-1,CNE-2Z,SUNE-1,SUNE-1-5-8F and SUNE-1-6-10B) in the levels of protein and mRNA.Methods The expression of Kiss-1 in the levels of protein and mRNA was detected by reverse transcription polymerase chain reaction(RT-PCR) and immunohistochemical staining technology.Results In human nasopharyngeal cell lines(CNE-1,CNE-2Z,SUNE-1,SUNE-1-5-8F and SUNE-1-6-10B) with the different metastasis ability,there was all positive expression of Kiss-1 protein and mRNA.The protein and mRNA levels of Kiss-1 highly expressed in human nasopharyngeal cell line(SUNE-1-6-10B) with low metastasis ability,lowly expressed in the other human nasopharyngeal cell lines(CNE-1,CNE-2Z,SUNE-1 and SUNE-1-5-8F) with high metastasis ability(P0.01).Conclusion The protein and mRNA of Kiss-1 were highly expressed in human nasopharyngeal cell line(SUNE-1-6-10B) with low metastasis ability,lowly expressed in the other human nasopharyngeal cell lines(CNE-1,CNE-2Z,SUNE-1 and SUNE-1-5-8F) with high metastasis ability.These indicated that Kiss-1 may play an important role in metastasis of nasopharyngeal carcinoma.
