摘要
目的建立鼻咽癌细胞肝转移亚株,研究鼻咽癌肝转移机制。方法利用裸鼠体内传代的方法获得肝转移细胞亚株,并用体内和体外实验来观察其侵袭转移能力。结果经体内反复筛选得到肝转移能力高的亚株5-8F-H3B-EGFP,其侵袭性、运动性以及形成肝转移灶的能力均大于母系5-8F-EGFP,并具有较高的肝脏亲和性。结论该亚株的建立为今后研究鼻咽癌肝转移的分子机制奠定了基础。
Objective To establish a hepatic metastatic subline of nasopharyngeal carcinoma (NPC) cell line. Methods NPC cells metastatic to the liver were isolated from nude mice and the invasion and metastatic ability of the cells was observed in vivo and in vitro. Results and Conclusion The invasion and metastasis activity of 5-8F-H3B-EGFP (an in vivo isolate with enhanced liver metastatic behaviors) were enhanced obviously in comparison with the parent cell line 5-8F-EGFP. This subline may be useful for cloning genes related to liver metastasis of NPC.
