摘要
目的:初步研究麦角甾醇(ergosterol,ES)对肝癌细胞HepG2增殖的影响及可能机制。方法:实验设立对照组和麦角甾醇10μg/ml、50μg/ml、100μg/ml、200μg/ml组,用MTT方法和CSFE染色流式细胞术观察麦角甾醇对HepG2细胞增殖的影响,RT-qPCR检测p21mRNA表达,Western blot检测p21蛋白的表达影响。结果:与对照组比较,作用48h、72h后,麦角甾醇50μg/ml、100μg/ml、200μg/ml均对HepG2有明显的抑制增殖作用,p21 mRNA和蛋白的表达量均明显增高。结论:麦角甾醇在50~200μg/ml可以抑制HepG2细胞增殖,其机制可能与增高p21mRNA和蛋白表达相关。
Objective: The present study observed the effect of ergosterol(ES) on proliferation of human liver cancer cell line HepG2,and investigated the potential mechanism.Methods: Five groups including control,ES 10μg/ml,ES 50μg/ml,ES 100μg/ml,ES 200μg/ml group were set up in this study.Cell proliferation was analyzed by MTT assay and by using flowcytometry with CSFE staining.p21 mRNA was evaluated by RT-PCR and real time PCR.Expression of p21 was detected by western blot.Results: Compared with the control,after treatment of ES of 50μg/ml,100μg/ml,200μg/ml for 24h,48h or 72h,HepG2 cell proliferation was inhibited,expression of p21 mRNA and protein both decreased.Conclusions: The above results indicated ES of 50μg/ml,100μg /ml,200μg /ml inhibited HepG2 cell proliferation by increased p21.
