熊果酸对糖尿病大鼠肾组织内核因子-κB活性及P选择素表达的影响 被引量:10

Effect of Ursolic Acid on the Activity of NF-κB and Expression of P-Selectin in Kidney of Diabetic Rats

摘要 目的:探讨熊果酸(UA)对糖尿病(DM)大鼠肾组织的保护作用及机制。方法:链脲佐菌素(STZ)诱导糖尿病大鼠模型,随机分为正常对照组(N组)、糖尿病组(D组)、糖尿病+UA治疗组(DM+UA组);EMSA法检测NF-κB活性,RT-PCR和Western blot分别检测P选择素mRNA和蛋白表达。结果:造模成功16周后,和D组相比,D+UA组尿蛋白排泄、肾功能和肾组织病理都得到一定程度改善,且UA治疗明显抑制了DM大鼠肾组织内NF-κB激活和P选择素表达。结论:熊果酸对DM大鼠肾组织具有一定的保护作用,其机制可能通过抑制NF-κB激活和P选择素表达相关。 Objective: To investigate the renal protection of ursolic acid(UA) in diabetic rats and its mechanism.Methods: Diabetic rats were induced by streptozotocin(STZ) and randomly divided into control group(N group),diabetic group(D group),and diabetes + UA group(D + UA group).NF-κB activity was detected by EMSA.The mRNA and protein levels of P-selectin expression were detected by RT-PCR and Western blot.Results: Sixteen weeks after diabetic rat modeling,the urinary protein excretion,renal function and renal pathology of rats in D + UA group had been improved to some extent as compared to D group.And UA treatment was significantly inhibited NF-κB activation and the expression of P selectin in renal tissue of diabetic rats.Conclusion: Ursolic acid has a protective effect,on renal tissue in diabetic rats,which may be associated with the inhibition of NF-κB activation and P selectin expression.
