保肝宁对实验性肝纤维化大鼠肝内核转录因子κB表达的影响

摘要 目的观察保肝宁对大鼠肝内核转录因子活性的影响。方法用CCl4造成大鼠慢性肝损伤模型,处理组给予不同剂量的保肝宁和秋水仙碱,病理组给予生理盐水,对照组为正常大鼠。应用免疫组化法观察肝脏细胞内NF-κB分布变化,Westemblot法观察肝脏细胞内磷酸化IkB的改变;HE染色观察肝组织病理改变。结果对照组肝脏细胞内NF-κB位于胞浆内,胞核内无表达,病理组NF-κB主要位于细胞核内,与病理组比较,保肝宁组明显抑制了肝脏细胞内NF-κB的核转位;保肝宁组IκB的磷酸化程度较病理组明显降低(P<0.01)。结论复方保肝宁明显抑制大鼠肝内NF-κB的表达,可能为其抗肝纤维化作用的机理之一。 Objective: To observe the effect of Herbal mixture Baoganning on expression of Nuclear factor - κappaB of fibrosio liver in rats. Methods: Rats liver fibrosis model was induced by carbon tetrachloride. The rats of control groups were treated with different dose of Herbal compund Baoganning and colchicines, respertirely. The rats of pathologic group were treated with Saline. The rats of contrast group were normal. The location of NF - κB in liver were detected Immunohistochemistty. The expression of IkB was detected by Western Blot. Results: The NF- κB of liver in pathology group located in the nuclear. But the NF- κB of liver in normal group located in cell plasm. Compared with the pathology group, the baoganning group inhibited the translocation of NF- κB to nuclear. The level of IκBα phosphorylation.in the liver of the treating group is significantly lower than in liver of the pathologic group( P 〈 0.01 ). Conclusion: Mixture Baoganning could significantly inhibit the expression of NF - κB, which is one of the mechanisms of mixture Baogarming treated hepatic fibrosis.
