摘要
目的观察藤茶提取物(FL0810)体内外抗肿瘤的作用。方法采用S180腹水瘤模型观察FL0810体内抗肿瘤作用。应用四甲基偶氮唑蓝(MTT)法研究FL0810对肿瘤细胞体外增殖的影响。结果 FL0810能延长S180腹水瘤小鼠生存时间,与模型对照组相比有显著差异(P<0.05);FL0810对人乳腺癌细胞MDA-MB-231、MDA-MB-435、人前列腺癌细胞DU145均有较强的体外增殖抑制作用,且呈现明显的剂量依赖性,抑制率随着药物浓度增加而增加;72 h半数抑制浓度(IC50)为别为208.71,226.14,150μg/mL。结论藤茶提取物在体内对S180的生长有一定的抑制作用,在体外对人乳腺癌和人前列腺癌有明显的抑制作用。
Objective To observe the anti-tumor effect of FL0810（an extract from Ampelopsis grossedentata） in vivo and in vitro.Methods S180 ascites tumor bearing mice model was used to observe inhibition of FL0810 on tumor growth in vivo.Then we applied methyl thiazolyl tetrazolium（MTT） method to study the effect of FL0810 on tumor cells proliferation in vitro.Results FL0810 could prolong the survival time of S180 ascites tumor bearing mice,and the difference was significant compared with the model control group（P 〈0.05）.FL0810 had obvious inhibition on in-vitro proliferation of human breast cancer cells MDA-MB-231,MDA-MB-435 and human prostate cancer cells DU145,and the effect was dose-dependent,inhibition rate increasing with drug concentration.IC50 after 72h for MDA-MB-231,MDA-MB-435 and DU145 was 208.71,226.14,150 μg/mL,respectively.Conclusion FL 0810 has certain inhibition on the growth of S180 in vivo,and has anti-tumor effect on human breast cancer cells and human prostate cancer cells in vitro.
