摘要
[目的]研究枸杞黄酮的提取工艺。[方法]采用不同的提取溶剂对枸杞叶及果柄中的黄酮进行提取,通过正交试验研究溶剂浓度、料液比、提取时间、原料粒度对总黄酮含量和得率的影响,确定枸杞黄酮的提取工艺。测定枸杞叶、果柄原料粗黄酮含量,分析不同样品中黄酮含量差异。[结果]确定了枸杞黄酮最佳提取工艺条件为:乙醇浓度70%,料液比1∶8(M∶V,g/ml),提取时间3 h,提取3~8mm碎粒,在该工艺条件下获得黄酮含量为79.4%,得率为3.72%。枸杞叶中黄酮含量明显高于枸杞果柄,5月份采摘的枸杞叶黄酮含量高于10月份采摘的。[结论]该研究为枸杞黄酮的开发提供了理论依据。
[Objective] To study the extraction technology of flavonoids from Lycium chinense Mill..[Method] The flavonoids was extracted from the leaves and fruit stalk of Lycium chinense Mill.by different solvents,the orthogonal experiment was adopted to study the effect of solvent concentration,solid-liquid ratio,extraction time and material partical size on the content and yield of total flavonoids,so as to determine the best extraction technology.The content of crude flavonoids in the leaves and fruit stalk was detected,and the difference of the flavonoids content in different samples was analyzed.[Result] The optimum extraction conditions were: 70% ethanol concentration,1∶8(M∶V,g/ml) of solid-liquid ratio,3 h extraction time and 3-8 mm broken granules,under those conditions the content and yield of flavonoids could reach 79.4% and 3.72% respectively.The flavonoids content in the leaves was obviously higher than that in the fruit stalk,and the wolfberry leaves picked in May had higher content of flavonoids than that picked in October.[Conclusion] The study provided theoretical basis for the development of Lycium chinense Mill.flavonoids.
