摘要
目的:从心肌组织胶原表达的角度观察益母草水提物对实验性大鼠心肌重构的影响。方法:SD大鼠皮下注射异丙肾上腺素建立心肌重构模型,随机分为正常对照组、模型组、依那普利组及益母草水提物低、高剂量组,药物干预15 w。检测大鼠心脏血流动力学指标、心输出量、心脏质量系数、左室质量系数、心肌羟脯氨酸含量、心肌组织Ⅰ、Ⅲ型胶原含量。结果:与模型组比较,益母草水提物高剂量能改善模型大鼠LVSP、+dp/dtmax和CO(P<0.05),益母草低剂量能改善-dp/dtmax、心脏质量系数和左室质量系数、心肌组织羟脯氨酸含量及Ⅰ、Ⅲ型胶原含量、Ⅰ/Ⅲ型胶原比(P<0.05)。结论:高剂量益母草水提物能改善ISO致大鼠心肌重构模型心脏收缩功能;低剂量益母草水提物能改善ISO致大鼠心肌重构模型舒张功能,下调胶原表达,改善心肌胶原构成比,减轻心肌重构程度。
Objective：To investigate the effects of Leonurus japonicas on myocardial remodeling induced by isoproterenol（ISO） in rats.Methods：The model of rat myocardial remodeling was established by subcutaneous injection of ISO（20,10 and 5 mg/kg for 3 days,subsequently 3 mg/kg for 7 days）.Rats were randomly divided into five groups：Control,ISO,ISO＋enalapril,ISO＋Leonurus 8g/kg and ISO＋Leonurus 16g/kg.Recorded the values of LVSP,LVEDP,±dp/dt_max,cardiac output（CO）.Weighed the body weight（BW）,heart weight（HW） and left ventricular weight（LVW） of rats to calculate the values of HW/BW and LVW/BW.The hydroxyproline contents were investigated by spectrophotometric measurement.The contents of typeⅠcollagen and type Ⅲ collagen and ratio ofⅠ/Ⅲ collagen were assessed by immunohistochemical stain.Results： Leonurus（16g/kg·day-1） could increased the value of LVSP,＋dp/dtmax,CO（P0.05）.Leonurus（8 g/kg） could increased the value of-dp/dt_max,decreased the ratios of HW/BW and LVW/BW（P0.05）,the contents of hydroxyproline,type Ⅰcollagen,type Ⅲ collagen and the ratios of Ⅰ/Ⅲ collagen（P0.05）.Conclusions：Leonurus at a dosage of 16 g/kg may improve the systolic function;Leonurus at a dosage of 8 g/kg may improve the diastolic function,down-regulate the expression of collagen and normalize the ratio of Ⅰ/Ⅲ collagen.
