摘要
目的:探讨新的小分子活性肽apelin对正常和低氧大鼠离体肠系膜小动脉环的舒张作用及其与一氧化氮(NO)途径的关系。方法:36只大鼠随机均分为正常组与低氧组,采用离体小血管环灌流法,观察apelin对去甲肾上腺素(NE)预收缩的大鼠离体肠系膜动脉环的舒张效应;L-Nω-硝基精氨酸甲酯(L-NAME)阻断后,观察apelin对NE预收缩的两组大鼠离体肠系膜动脉环的舒张效应。结果:①apelin对正常组和低氧组肠系膜小动脉均有舒张效应(P<0.05)。②apelin对低氧组的舒张效应大于正常组(P<0.05)。③L-NAME阻断后,apelin的舒张效应明显减弱,但正常组和低氧组差异无统计学意义(P>0.05)。结论:apelin对正常和低氧大鼠离体肠系膜小动脉均有舒张作用;apelin舒张肠系膜小动脉环的作用部分是通过一氧化氮途径实现的。
Objective: To investigate the diastolic effect of apelin on mesenteric artery ring and its relationship with nitric oxide(NO) pathway in normal and hypoxic rats.Methods: Thirty-six male Sprague-Dawley rats were randomly divided into normal control group and hypoxia group.The mesenteric arteriolar diastolic activity in vitro was used.After norepinephrine(NE) pre-contraction,apelin relaxed the mesenteric artery ring in a dose-dependent manner.The relaxation effect of apelin was significantly attenuated in the presence of L-NAME.Results: Apelin had significant relaxation effects on mesenteric arterial rings in hypoxia group and normal control group(P 0.05).The relaxation effect of apelin in hypoxia group was heavier than normal control group(P 0.05).Apelin had no significant effects between normal control group and hypoxia group with L-NAME(P 0.05).Conclusion: Apelin has significant relaxation effect on mesenteric arterial rings in hypoxia group and normal control group,the hypoxia group is more significant,and its mechnism is partly through of nitric oxide pathway.
