摘要
目的 建立一种人肠系膜小动脉平滑肌细胞急性酶分离法并在分离的细胞上研究其离子通道特性 .方法 采用木瓜蛋白酶、胶原酶和透明质酸酶分离出人肠系膜小动脉平滑肌细胞 ,用膜片钳全细胞记录法记录通道电流 .结果 该方法分离过程简单 ,所获细胞数量多 ,形态正常 ,在这些细胞上容易记录到正常的外向钾通道电流 .结论 应用本方法易得单个活性正常。
Objective To establish an enzymatic isolation method of human mesenteric arteriolar smooth muscle cells and study ion channel characteristics of the isolated cells. Methods Papain,collagenase and hyaluronidase were used to isolate single human mesenteric arteriolar smooth muscle cells and whole-cell recording techniques of patch clamp was used to record channel current. Results The isolation process was convenient.Many isolated cells with normal configuration of membrance were obtained. Conclusions Single human mesenteric arteriolar smooth muscle cells with normal activity and whole configuration were easily ganied with the method.
