摘要
[目的]建立并完善石榴叶总黄酮提取和含量测定方法,筛选出最佳提取工艺条件。[方法]以芦丁为对照品,采用分光光度法在最大吸收波长510 nm处对其含量进行测定。[结果]测定石榴叶中的总黄酮含量为0.774%(0.774 mg/g)。最佳提取工艺为乙醇浓度70%、料液比1∶50(g/ml)、超声提取时间为30 min。[结论]选用芦丁为对照品应用于分光光度法测定石榴叶总黄酮含量,方法简单、准确度高、用时短,可用于石榴叶总黄酮的含量测定。
[Objective]The aim is to extract and determine total flavonoids from from the leaves of Punica granatun L.,that select the best extract industry item.[Method] With rutin being taken as the standard sample,determined the flavonoids in the sample by spectrophotometry at 510 nm.[Result] In the detected samples,that total flavonoids concentration C=0.774%(0.774 mg/g),the best extract industry: methanol concentration is 70%,solid liquid ratio is 1∶50,extraction time is 30 min.[Conclusion] Rutin is selected as reference substance to determine content of total flavonoids in the leaves of Punica granatun L.with spectrophotometry,method is simple,degree of accurancy is high and the time is shorter,that can determine the flavonone content in the leaves of Punica granatun L..
