摘要
以利用复活促进因子(Rpf)从土壤和污水处理系统中分离得到的菌种作为筛选絮凝剂产生菌的菌源,构建出产絮凝剂的复合菌群M3和M7.通过单因素实验,以絮凝率(FE)和菌体生长(OD660)为评价指标,对培养基的组成及培养条件进行优化研究.结果表明:该复合菌群M3和M7在组分为淀粉1.5%,(NH4)2SO40.3%,FeCl20.1%,pH 7.0的发酵培养基上,经预发酵,以复配比(M3∶M7)为0.6∶0.4,6%(V/V)的总接种量,28℃,160r/min发酵培养72h后,其产生的高效絮凝剂对高岭土悬浊液的絮凝率达96.27%.
Using bacteria isolated from soil and sewage treatment system by adding resuscitation promoting factor (Rpf) to culture medium as source, We constructed two bioflocculant-producing bacterium M3 and M7. Through measuring the flocculating efficiency (FE) and the cell growth (OD660), the optimal medium compositions and culture conditions of multiple bioflocculant-producing microorganisms were studied by single factor. The results showed that the optimal conditions medium compositions were starch 1.5%, (NH4)23O4 0.3%, FeCl2 0.1%, initial pH 7.0; The optimal conditions for bioflocculant production were : the proportion of M3 to M7 0.6 : 0.4, the total inoculum size 6% (V/V), fermentation temperature 28℃, fermentation time 72 h and shaking speed 160 r/min. Under these conditions, the flocculating efficiency reached 96.27%.
