摘要
以苦瓜叶为原料,采用复合酶法从苦瓜叶中提取黄酮。通过对单一酶和复合酶辅助提取的实验效果比较,得到了纤维素酶和果胶酶组成的复合酶为最佳复合酶,通过单因素试验和正交试验确定复合酶提取苦瓜叶中总黄酮的最佳工艺条件:纤维素酶浓度为0.8mg/mL,果胶酶浓度为0.5mg/mL的情况下,提取温度为55℃、提取介质pH4.5的条件下酶解140min,苦瓜叶总黄酮的提取率可达到3.13%,与直接醇提工艺相比较,总黄酮提取率提高了30%以上。对苦瓜叶中黄酮有效成分进行提取研究,为合理有效利用苦瓜叶这一废弃物提供参考依据,具有重要的理论意义和实际应用价值。
Taking leaves of Momordica Charantia L.as material,and extracting the total flavonoids from it by compound enzymes.The catalytic effect was compared between single enzymes and compound enzymes,the result showed that the best composition of compound enzymes was cellulase and pectolase.Based on the single factor tests and orthogonal experiments,the optimum conditions by compound enzymatic treatment were as follow:amount of compound enzyme were 0.8mg/mL cellulase and 0.5mg/mL pectinase,enzymatic temperature 55℃,enzymatic treatment time for 140 min and pH value 5.0.On this condition,the extraction rate of flavonoids could get up to 3.13%.Compared with extraction of alcohol,the extraction rate was increased by over 30%.Study on extracting the total flavonoids from the leaves of Momordica Charantia L.it provide reference information for reasonable and effective use,have important theoretical and practical significance.
