摘要
目的:探讨肿瘤坏死因子相关凋亡诱导配体(TRAIL)信号途径在多囊卵巢综合征(PCOS)大鼠卵巢颗粒细胞诱导凋亡的最终效应阶段的作用。方法:采用硫酸普拉睾酮钠诱导大鼠PCOS模型,采用Western blot及RT-PCR分析半胱氨酸天门冬氨酸蛋白酶3(Caspase3)在PCOS大鼠卵巢颗粒细胞的表达情况。结果:①Western blot分析显示,PCOS组大鼠卵巢颗粒细胞活性Caspase3蛋白的表达较对照组明显增强(P<0.01)。②RT-PCR分析显示,PCOS组大鼠卵巢颗粒细胞Caspase3mRNA的表达较对照组明显增强(P<0.01)。结论:TRAIL信号途径在PCOS大鼠颗粒细胞诱导的凋亡可能最终经过了Caspase级联反应,引起Caspase3的激活导致细胞凋亡。
Objective:To explore the role of tumor necrosis factor-related apoptosis-inducing ligand(TRAIL) signal pathway in the final effective stage of apoptosis of granulosa cells of rats with polycystic ovary syndrome(PCOS). Methods:Sodium prasterone sulfate was used to induce rat models of PCOS,Western blot and RT-PCR were used to analyze the expression of Caspase3 in granulosa cells of rats with PCOS. Results:Western blot analysis showed that the expression level of actived Caspase3 protein in PCOS group was significantly higher than that in control group(P0.01).RT-PCR analysis showed that the expression level of Caspase3 mRNA in PCOS group was significantly higher than that in control group(P0.01). Conclusion:TRAIL signal pathway may pass Caspase cascade finally during the apoptosis of granulosa cells of rats with PCOS,induce cell apoptosis by activating Caspase3.
