摘要
目的：观察金匮温经汤对围绝经期大鼠血清雌二醇（E2）、卵泡刺激素（FSH）、黄体生成素（LH）及卵巢凋亡相关蛋白Caspase3、Caspase9、Apaf-1达的影响,从细胞凋亡角度探讨金匮温经汤治疗围绝经期综合征的作用靶点。方法：12月龄SD雌性清洁级大鼠,采用阴道脱落细胞涂片法选取40只围绝经期模型大鼠,随机数字表法分为模型组,中药组（各20只）,对照组为20只3月龄SD雌性大鼠。中药组给予金匮温经汤8. 31 g·kg^-1·d^-1灌胃,模型组、对照组均给予同体积蒸馏水灌胃,每日1次,连续28 d。处死大鼠,采用放免法观察大鼠血清E2、FSH、LH的水平;取卵巢,采用Western Blot检测凋亡相关蛋白Caspase3、Caspase9、Apaf-1表达。结果：模型组,血清E2水平低于对照组（P 〈0. 05）,FSH、LH高于对照组（P 〈0. 05）;卵巢Caspase3、Caspase9、Apaf-1蛋白表达均高于对照组（P 〈0. 05）。中药组,血清E2水平高于模型组（P 〈0. 05）,FSH、LH低于模型组（P 〈0. 05）;卵巢Caspase3、Caspase9、Apaf-1蛋白表达下调,均低于模型组（P 〈0. 05）。结论：金匮温经汤通过下调卵巢凋亡因子Caspase3、Caspase9、Apaf-1蛋白表达,以此改善生殖内分泌水平,治疗围绝经期综合征。
Objective： To observe the effects of Jingui Wenjing Decoction on serum estradiol（ E2）,follicle stimulating hormone（ FSH）,luteinizing hormone（ LH） and ovarian apoptosis related proteins Caspase3,Caspase9 and Apaf-1. To explore the role of Jingui Wenjing Decoction in the treatment of perimenopausal syndrome from the perspective of apoptosis. Methods： Forty SD female rats which were 12 months old were randomly divided into model group and traditional Chinese medicine group（ n = 20） by using vaginal exfoliated cell smear method. The control group had 20 SD female rats which were 3 months old. The traditional Chinese medicine group was fed with Jingui Wenjing Decoction 8. 31 g/（ kg·d）. The model group and the control group were given the same volume of distilled water,once a day for 28 days. The rats were sacrificed and the levels of serum E2,FSH and LH were detected by radioimmunoassay. The ovarian tissues were taken out and the expressions of Caspase3,Caspase9 and Apaf-1 were detected by Western blot. Results： The levels of E2 of the model group were lower than those of the control group（ P〈0. 05）. FSH and LH were higher than those of the control group（ P〈0. 05）,while the expressions of Caspase3,Caspase9 and Apaf-1 were higher than those of the control group. The level of E2 of Chinese herbal medicine group was higher than that of the model group（ P〈0. 05）.FSH and LH were lower than those of the model group（ P〈0. 05）,while the expressions of Caspase3,Caspase9 and Apaf-1 were lower than those of the model group. Conclusion： Jingui Wenjing Decoction can reduce the levels of ovarian apoptosis factors Caspase3,Caspase9 and Apaf-1 in order to improve reproductive and endocrine levels and treat perimenopausal syndrome.
