摘要
目的:采用比较蛋白质组学方法研究良性转归葡萄胎和恶性转变葡萄胎中的差异表达蛋白,筛选葡萄胎恶变相关蛋白。方法:分别提取3例良性转归和3例恶性转变完全性葡萄胎组织,进行双向凝胶电泳,质谱鉴定差异蛋白,免疫组化方法分析差异蛋白氯离子通道蛋白1(CLIC1)在葡萄胎和绒毛膜癌中的相对表达量。结果:通过二维电泳分离蛋白和差异比对,共筛选到34个差异蛋白点,经过质谱技术共鉴定出18个差异蛋白。生物信息学分析表明绝大多数差异蛋白参与细胞增殖和细胞存活过程。挑选了一个差异蛋白CLIC1蛋白进行进一步的研究,结果表明绒癌组织中CLIC1明显高于完全性葡萄胎组织,P<0.01。恶性转变葡萄胎组织中CLIC1表达也明显高于良性转归葡萄胎,P<0.01。结论:通过蛋白质组学CLIC1筛选结果表明,CLIC1蛋白可能成为葡萄胎恶性转变的预测指标。
OBJECTIVE: To identify some prognostic biomarkers for the malignant transformation of hydatidiform moles (HMs). METHOIIS: 2-DE based proteomic approach was used to compare the protein profiles of complete benign moles (3 samples) with those of malignant-transformed moles (3 samples). MALDI-TOF-TOF was used to identify the differential proteins. Western blot was used to verify the results of 2DE and Immunohistology was used to explore the function of differential protein in gestational trophoblastic disease (GTD). RESULTS.. Thirty-four protein points were selected. Eighteen proteins deregulated in the maligrmnt-transformed group compared with the benign group were identified. Bioinformatic analysis indicated that most of these 18 proteins were involved in the processes of cell proliferation and cell survival. Among the 18 proteins, chloride intracellular channel protein 1 (CLIC1), was chosen for further study. It showed that the levels of CLIC1 expression in choriocarcinoma tissue were higher than that in complete HM tissue (P〈0.01). CLIC1 expression was increased in the tissues of malignant-transformed HMs compared with non- transformed HMs (P〈0. 01). CONCLUSION: Our findings confirm the potential of CLIC1 may be a prognostic marker for the malignant transformation of HMs.
