摘要
目的:建立同时测定大卫颗粒中绿原酸、连翘苷、黄芩苷的高效液相色谱法。方法:采用Sinochrom ODS-BP(250mm×4.6mm,5μm)色谱柱,流动相为甲醇:30mmol·L^(-1)磷酸二氢钾溶液,梯度洗脱,流速为1.0ml·min^(-1),检测波长为277nm。结果:绿原酸在10~80μg·ml^(-1)范围内线性关系良好(r=0.9996),平均回收率为98.85%,RSD=1.02%(n=9);连翘苷在10~80μg·ml^(-1)范围内线性关系良好(r=0.9993),平均回收率为99.17%,RSD=0.99%(n=9);黄芩苷50~600μg·ml^(-1)范围内线性关系良好(r=0.9995),平均回收率为98.72%,RSD=1.10%(n=9)。结论:本方法简便易行,能够准确、快速测定大卫颗粒中的绿原酸、连翘苷和黄芩苷的含量。
To establish an HPLC method for determining chlorogenic acid, phillyrin and baicalin in Dawei granules. Method: The separation column of Sinochrom ODS-BP (250 mm ×4. 6 ram,5 μm) was used,methanol-30 mmol·L^-1,K2HPO4 was applied as the mobile phase with gradient elution, the column temperature was 40 ℃, the detection wavelength was 277 nm and the flow rate was 1.0 ml·min ^-1. Result: The linear range of chlorogenic acid was 10-80μg. ml^-1 ( r = 0. 999 6) with the average recovery of 98.85% ( RSD = 1.02%, n = 9 ), that of phillyrin was 10-80 μg·ml ^-1( r = 0. 999 3 ) with the average recovery of 99. 17% ( RSD = 0. 99% n = 9 ), and that of baicalin was 50-600μg. ml^-1 ( r = 0. 999 5 ) with the average recovery of 98. 72% ( RSD = 1.10% n = 9 ). Conclusion: The method is simple, rapid and sensitive, which is suitable for the content determination of chlorogenic acid,phillyrin and baicalin in Dawei granules.
