摘要
目的建立HPLC-DAD法同时测定小儿肺热咳喘口服液(麻黄、甘草、金银花、连翘、黄芩、苦杏仁等)中9个成分(盐酸麻黄碱、盐酸伪麻黄碱、绿原酸、连翘酯苷B、甘草苷、木犀草苷、黄芩苷、连翘苷、甘草酸)。方法采用HPLC-DAD方法,ZORBAX SB-C18色谱柱(4.6 mm×250 mm,5μm);流动相为乙腈(A)-0.1%磷酸溶液(B)梯度洗脱,体积流量0.8 mL/min,柱温为35℃,检测波长为210 nm(盐酸麻黄碱、盐酸伪麻黄碱、连翘酯苷B、甘草苷、连翘苷),253 nm(甘草酸),278 nm(黄芩苷),327 nm(绿原酸),350 nm(木犀草苷)。结果 9个成分的峰面积与质量浓度的线性关系良好,加样回收率为95.6%~99.6%。结论该方法简便、准确、灵敏,重复性好,可用于小儿肺热咳喘口服液中多种指标性成分的质量控制。
AIM To develop an HPLC-DAD method for determining nine constituents,ephedrine hydrochloride,pseudoephedrine hydrochloride,chlorogenic acid,forsythiaside B,liquiritin,luteolin-7-O-glucoside,baicalin,phillyrin and glycyrrhizic acid in Xiao′er Feire Kechuan Liquid(Ephedrae Herba,Glycyrrhizae Radix et Rhizoma,Lonicerae japonicae Flos,Forsythiae Fructus,Scutellariae Radix,Armeniacae Semen amarum,etc.).METHODS Nine constituents were analyzed by ZORBAX SB-C18(4.6 mm×250 mm,5 μm)with acetonitrile-water(containing 0.1% phosphonic acid) as mobile phases in gradient elution,0.8 mL/min as flow-rate and 35 ℃ as temperature of column.The detection wavelengths were at 210 nm for ephedrine hydrochloride,pseudoephedrine hydrochloride,forsythiaside B,phillyrin,253 nm for glycyrrhizic acid,278 nm for baicalin,327 nm for chlorogenic acid and 350 nm for luteolin-7-O-glucoside,respectively.RESULTS Nine constituents were seperated perfectly and good linear relationships were obtained.The recoveries were between 95.6%-99.6%.CONCLUSION The developed method is simple,specific,accurate and with good reproducibility,which can be used for quality control of Xiao′er Feire Kechuan Liquid.
