摘要
目的检测胃癌组织中中期因子(MK)、尿激酶型纤溶酶原激活物(UPA)的表达,探讨MK、UPA与胃癌的侵袭、转移的关系,并与MK、UPA的相关性进行分析。方法应用免疫组织化学两步法检测60例胃癌组织(胃癌组)中MK、UPA的表达,并以20例胃癌旁(距离肿瘤边缘5 cm)的正常组织标本作为对照组。结果胃癌组织中的MK、UPA的表达率分别为63.33%、51.67%,对照组的MK、UPA的表达率分别为25.00%、10.00%,2组比较差异有统计学意义(P<0.05)。浸润全层的MK、UPA的表达高于浸润黏膜层、肌层的胃癌(P<0.05);低分化的MK、UPA的表达高于中分化胃癌(P<0.05);有淋巴结转移的MK、UPA的表达高于无淋巴结转移胃癌(P<0.05)。MK与UPA的表达呈正相关(r=0.579,P<0.001)。结论 MK、UPA的表达与胃癌的侵袭、转移密切相关,二者联合检测可作为判断胃癌发展及预后的分子指标。
Objective To detect the expression of midkine（MK） and urokinase-type plasminogen activator（UPA） in human gastric cancer,and to investigate the relationship of MK and UPA expression to the invasion and metastasis of gastric cancer.Methods The expression of MK and UPA was detected by immunohistochemistry in 60 cases of gastric cancer and 20 cases of normal control tissue（5 cm from the margin of the tumor）.Results The expression rates of MK and UPA in cancer tissue were significantly higher than those in normal control tissue（63.33% vs 25.00% and 51.67% vs 10.00%,respectively,P0.05）.The expression of MK and UPA in gastric cancer with full-thickness infiltration was higher than that in gastric cancer with infiltration of mucosa and muscular layer（P0.05）.The expression of MK and UPA in poorly differentiated gastric cancer was higher than that in well-differentiated gastric cancer（P0.05）.The expression of MK and UPA in gastric cancer with lymph node metastasis was higher than that in gastric cancer without lymph node metastasis（P0.05）.There was a positive correlation between MK and UPA expression（r=0.579,P0.001）.Conclusion The expression of MK and UPA was closely related to the invasion and metastasis of gastric cancer,and MK expression was positively correlated with UPA expression.A combined detection of MK and UPA may be used to determine the development and prognosis of gastric cancer.
