摘要
以榆叶梅花粉为试材,利用离体培养法、碘-碘化钾染色法、醋酸洋红染色法和TTC(氯化三苯基四氮唑)染色法对花粉生活力进行测定,并研究了不同贮藏条件对花粉萌发的影响。结果表明:培养基中添加不同浓度的蔗糖和硼酸对花粉的萌发有促进作用,榆叶梅花粉萌发的最适条件为在培养基中添加15%蔗糖+0.004%硼酸,萌发率达41.52%。花粉生活力随着贮藏时间的延长而下降,不同的贮藏条件对其有显著的影响,适当的低温可有效延长花粉贮藏时间。其中-80℃萌发率最高,为最佳贮藏方法。
Take the fresh pollen of Prunus triloba as experiment material,germination in vitro,I2-KI staining,carmine acetate dyeing and TTC staining methods were used to determine the fresh pollen ability of Prunus triloba.Effects of different storage on the pollen germination were also studied.The results showed that culture media with different concentration of sucrose and boric had good effects on the pollen germination.The pollen germinating percentage reached the highest after incubated on the media supplemented with 15% sucrose and 0.004% Boric acid,the germination of pollen reached 41.52%.Vitality of pollen were gradually descend as last of store time,the suitable low temperature effective extension the store time of pollen.The best methods to keep vitality of pollen at-80℃,the germination of pollen is higher.
