摘要 【目的】掌握中华蜜蜂蜂毒明肽基因的生物信息,为进一步深入研究其生理功能及通过生物工程技术生产蜂毒明肽产品奠定基础。【方法】采用PCR技术克隆中华蜜蜂蜂毒明肽基因,并对所获得的基因组序列进行详细的生物信息学分析。【结果】从蜜蜂头部和胸部组织中成功扩增获得511bp的蜂毒明肽基因序列,该序列GC碱基含量仅为24%,包含一个141bp的开放阅读框,编码46个氨基酸残基;该序列与肥大细胞脱颗粒肽的同源性高达58%,存在较明显的信号肽序列和跨膜区结构。【结论】蜂毒明肽是一种小分子抗菌肽,存在较明显的信号肽序列和跨膜区结构,具有生物抗菌肽的典型特性,但需进一步加工切除信号肽序列后才能发挥作用。 [Objective]The present experiment was conducted to isolate the apamin gene from Apis cerana cerana in order to characterize its properties and to provide information for understanding its physiological functions. [Method]Apamin gene was isolated from head and chest tissues of A. cerana cerana, cloned and sequenced for nucleotides and amino acids. [Result]The amplified apamin gene from head and chest tissue of A. cerana cerana was 511 bp in length with 24% GC content. The sequence contained a 141 bp open reading frame which encoded 46 amino acid residues. The protein of apamin showed 58% homology to mast cell degranulating-peptide precursor, which is another important bioactive peptide in bee toxin. A signal peptide sequence and transmembrane domain were also found in apamin protein. [Conclusion]The apamin is a micromolecule antibiosis peptide, and has a signal peptide sequence and transmembrane domains and characteristics of bio-antibiosis peptide. It may play an important role after deleting the signal peptide sequences.
