摘要
目的:观察氯沙坦对动脉粥样硬化程度和黏附分子的影响。方法:将24只雌雄各半新西兰白兔随机均分为对照组、高脂组、治疗组。对照组喂普通饲料,高脂组喂以高脂饲料,治疗组为高脂饲料加用氯沙坦25mg/(kg.d)。分别于实验开始前、开始后第4、8、12周清晨空腹取各组实验动物耳中央静脉血0.5ml,分别测定并比较各组空腹状态下ICAM-1、VCAM-1。于第12周末放血法处死全部动物。将两侧颈动脉取出,苏丹Ⅲ染色测主动脉斑块面积,HE染色观察。结果:实验后不同时间点各组间ICAM-1、VCAM-1水平的比较显示,高脂组高于对照组,治疗组均较高脂组降低,差异有统计学意义(P<0.01)。主动脉斑块面积治疗组明显<高脂组,差异有统计学意义。HE染色结果分析,高脂组内膜明显增厚,治疗组较高脂组减轻。结论:氯沙坦明显抑制颈动脉硬化斑块的产生并能显著降低黏附分子的表达。
Objective To study the effects of losartan on carotid atherosclerosis,serum intercellular adhesion molecule-1(ICAM-1) and vascular cell adhesion molecule-1(VCAM-1).Methods Twenty-four New Zealand rabbits were randomly and equally assigned to three groups: control group fed with normal diet,high-cholesterol group fed with high-cholesterol diet alone and Losartan-treated group fed with high-cholesterol diet and treated with Losartan 25mg/(kg·d).0.5ml blood from central veins of the ears in the morning was collected before the experiment and at 4,8,and 12 wks after the experiment respectively.The animals were fasted for 12 hours before the blood collection.Serum ICAM-1 and VCAM-1 were measured with ELISA.All the rabbits were killed by depletion method after 12 wks.Bilateral carotids were separated.The area of CAP with sudan III staining and the tunica media with hematoxylin-eosin staining were measured.Results The levels of serum ICAM-1 and VCAM-1 in high-cholesterol group were higher than control group at each timepoint after the experiment.Compared with high-cholesterol group,the levels of serum ICAM-1 and VCAM-1 in treatment group were significantly lower 8,and 12 wks(P0.01).The atherosclerotic plaque was less obvious in treatment group than which in high-cholesterol group.Observed with hematoxylin-eosin staining,the tunica intima thickened in high-cholesterol group and which in treatment group was less obvious.Conclusion Losartan could significantly inhibit the development of the carotid artery atherosclerosis and prominently reduce the expression of serum ICAM-1 and VCAM-1.
