摘要
目的探讨大鼠马尾神经受压后相应节段脊髓神经细胞中Bcl-2、Bax蛋白的表达以及Bcl-2/Bax与神经细胞凋亡的相关性。方法 30只健康成年SD大鼠随机分为对照组和实验组。对照组仅切除L5椎体,实验组切除L5椎体后在L4椎体平面置入硅胶片,造成马尾神经受压。实验组分别于建模后第2、4、8、12周取L4节段脊髓组织,每个时间点6只,对照组在手术后第8周取材。应用免疫组织化学法检测Bcl-2和Bax蛋白表达,采用原位末端标记法标记凋亡细胞,透射电镜观察脊髓神经细胞形态。结果对照组Bcl-2、Bax的OD值均为0.13±0.01。实验组从第2周到第12周Bcl-2的OD值分别为0.15±0.02、0.23±0.02、0.29±0.07、0.30±0.04,Bax的OD值分别为0.15±0.02、0.30±0.02、0.48±0.02、0.47±0.10。实验组从第4周开始Bcl-2、Bax表达明显增多。对照组及实验组第2、4、8、12周Bcl-2/Bax的比值分别为0.99±0.29、0.96±0.05、0.75±0.04、0.61±0.07、0.65±0.07,对照组和实验组第2、4、8、12周TUNEL染色阳性的凋亡细胞数分别为1.08±0.11、1.09±0.10、3.30±0.16、4.45±0.14、4.51±0.17。Bcl-2/Bax比值与凋亡细胞数量的相关系数为-0.891(P<0.05)。透射电镜观察发现,实验组在4、8、12周时脊髓神经细胞出现凋亡改变。结论马尾神经慢性受压后Bcl-2、Bax蛋白表达增加,Bcl-2/Bax与神经细胞凋亡呈负相关,提示Bcl-2、Bax对马尾神经受压后脊髓神经细胞的凋亡可能具有一定的作用。
Objective To observe the expression of Bcl-2, Bax in neurons of the spinal cord 'after chronic cauda equina compression and the correlation of Bcl-2/Bax with apoptosis of the neurons. Methods 30 healthy adult SD rats were randomly divided into control group and experimental group. In the control group, only the centmm L5 was resected. In the experimental group, after the resection of L5, a silicon sheet was inserted into the spinal canal at the L4 level to cause compression of cauda equina. L4 level of spinal cords were harvested in the 2nd, the 4th, the 8th and the 12th week after operation in the experimental group and in the 8th week in the control group respectively. Six were harvested each time. Then the expression of Bcl-2 and Bax were detected by using irmnunohistochemistry method and apoptotie cells were marked by TUNEL staining. The positive cells of Bcl-2 and Bax and neuron apoptosis were observed by light microscope and analyzed by computer image analysis system. The morphology of cells was observed by transmission electron microscope. Results The OD values of Bcl-2 and Bax were 0.13±0.01 in the control group. The OD values of Bcl-2 were 0.15±0.02(2nd week),0.23±0.02(4th week),0.29±0.07 (8th week) and 0.30±0.04 ( 12th week)respectively in the experimental group. The OD values of Bax were 0.15±0.02 (2nd week),0.30± 0.02(4th week ), 0.48±0.02 (8th week ) and 0.47±0.10, (12th week) respectively in the experimental group. The expression of Bcl-2 and Bax increased obviously from the 4th week. The ratios of Bcl-2/Bax were 0.99±0.29 (0 week),0.96±0.05 (2nd week),0.75±0.04(4th week), 0.61±0.07 (8th week), 0.65±0.07 (12th week ) respectively. The number of apoptotic cell was 1.08±0.11 in the control group and 1.09±0.10 ( 2nd week ), 3.30±0.16 (4th week ), 4.45±0.14 ( 8th week ) and 4.51 ±0.17 ( 12th week ) in the experimental group respectively.
