摘要
研究了节节草总生物碱的提取方法及抑菌活性。通过单因素试验和正交试验确定了最佳提取条件,测定了提取物对奇异变形杆菌、大肠杆菌、藤黄微球菌、黑曲霉菌、枯草杆菌、产气杆菌、青霉菌、金黄葡萄球菌的体外最小抑菌浓度及抑菌圈直径。结果表明,最佳工艺条件如下:盐酸浓度为1.25%、固液比为1∶18、在90℃的水浴中提取1 h、提取次数为3次;此条件下,节节草总生物碱提取率为9.5%;除青霉菌、金黄葡萄球菌外,提取物对其他菌的抑菌圈直径均≥7 mm;提取物对大肠杆菌、藤黄微球菌的最小抑菌浓度最低,为0.16 mg/mL;对奇异变形杆菌、大肠杆菌、藤黄微球菌、黑曲霉菌均具有强的抑菌作用。
The extraction methods and antibacterial activity of total alkaloids from Equisetum hiemale L.were studied.The single factor experiment and orthogonal test were conducted to find out the optimal extracting conditions and determine the minimum in vitro inhibitory concentration(MIC) and diameter of inhibition zone of total alkaloids extracted from Equisetum hiemale L.against Proteus mirabilis,Escherichia coli,Micrococcus luteus,Aspergillus niger,Bacillus subtilis,Bacillus aerogenes,Penicillium and Staphylococcus aureus.The results showed that 9.5% total alkaloids can be obtained from Equisetum hiemale L.when extraction conditions were as follows: 1.25% HCl,1∶18 of solid-liquid ratio,90℃ of water bath temperature,1 h of extraction time and repeating 3 times;except Penicillium and Staphylococcus aureus,the diameter of inhibition zone of total alkaloids on the other bacteria were over 7 mm;the MIC of total alkaloids on Escherichia coli and Micrococcus luteus was the lowest by 0.16 mg/mL;and the total alkaloids had strong antibacterial effect on Proteus mirabilis,Escherichia coli,Micrococcus luteus and Aspergillus niger.
