酶法提取杏鲍菇多糖及抗氧化活性研究 被引量:15

Research on Extracted Methods and Anti-oxidative Activity of Pleurotus eryngii Polysaccharide by Enzymes Method

摘要 以杏鲍菇为原料,采用水提和复合酶法提取杏鲍菇多糖,用苯酚-硫酸法测定多糖含量,探讨提取工艺条件,用Sevag法脱蛋白对杏鲍菇多糖进行纯化,并对其抗氧化活性进行研究。试验结果表明,水提最佳条件为料液比1∶20(g/m L)、时间50 min、温度30℃,多糖提取率为13.64%;酶法提取最佳工艺条件为酸性纤维素酶2.0%、酸性蛋白酶1.5%、料液比1∶20(g/m L)、温度30℃、时间50 min,多糖提取率为15.86%;复合酶法比水提法提取率提高了16.28%。采用Sevag法对杏鲍菇粗多糖进行纯化,多糖纯度提高了40.37%。杏鲍菇粗多糖,精多糖对OH·、O2-·和DPPH均有较强的清除能力,且随其浓度的增加清除率逐渐增大;相同质量浓度粗多糖的清除效果大于精多糖,但都明显低于VC。 Pleurotus eryng/i polysaecharide were extracted by water-complex enzymes methods and detected by phenol-sulphuric acid method. Deproteinization method of Sevag and anti-oxidative activity were discussed. The results showed that the best extracted conditions of water extracted method was solid/liquid 1 : 20 (g/mL), 30 % for 50 min with 13.64% extraction ratio ofpolysaccharide. The best extracted conditions of enzymes method was 2.0% acid cellulase, 1.5% acid protease, solid/liquid 1 : 20 (g/mL), 30 ℃ for 50 min with 15.86% exlracfion ratio of polysaccharide. Extraction ratio of complex enzymes method was 16.28% higher than water exlracted method Polysaccharide purity of Pleurotus eryngii raised 40.37% after depmteinization. Both crude and fine Pleurotus eryng/i polysaccharide had strong clearance power of OH ·, · 02 and DPPH. The clearance rate added with increasing polysaccharide concentration. The clearance rate of crude polysaccharide was higher than that of fine polysaccharide at the same concentration. Both of clearance rates were lower significantly than that of VC.
