摘要
为优化‘砀山酥梨’转基因试管苗的生根条件,解决梨试管苗生根的难题,以‘砀山酥梨’转基因试管苗作为试验材料,研究了基本培养基类型、培养方式、不同的间苯三酚(PG)浓度、不同的IBA浓度和暗培养时间对生根的影响。结果表明:采用两步生根法,试管苗在附加80 mg/L PG和20 mg/L IBA的ASH基本培养基上暗培养7d后转入附加80 mg/L PG的ASH培养基上进行光照培养,生根率达到91.7%。
To optimize the conditions of rooting of in vitro transgenic Pyrus bretschneideri Rehd.cv.Dangshansuli,in the present study,the effects on the rooting of in vitro transgenic Pyrus bretschneideri Rehd.cv.Dangshansuli were assessed using different basic media,rooting method,dark culture time,and concentrations of Phloroglucinol(PG) and IBA.The results indicated that the best induction root was achieved by a two-step rooting method:firstly the plantlets in vitro were incubated on ASH medium with 80 mg/L PG and 20 mg/L IBA in the dark for 7 days;and then the shoots were transferred to the same medium with 80 mg/L PG in the normal light conditions.As a result,91.7% of the rooting rate was obtained under above conditions.
