摘要
目的:采用大鼠急性脊髓损伤模型,观察电针对脊髓损伤后神经细胞凋亡与其相关基因Caspase-3表达及变化规律。试图从线粒体启动凋亡途径相关主要因素的研究角度进一步揭示电针治疗脊髓损伤作用机制。方法:采用A llen's法制备急性脊髓损伤模型。动物分为电针组、药物组(Caspase-3抑制剂组)、模型组、假手术组。应用Tunel法对细胞凋亡进行标记。结果:药物组治疗结果表明,Caspase-3在3天,药物组表达量低于模型组(P<0.05);电针组治疗结果表明,Caspase-3在14天,电针组表达量低于模型组(P<0.05)。结论:大鼠急性脊髓损伤后存在细胞凋亡现象,是脊髓继发性损伤主要病理机制。Caspase-3在大鼠脊髓损伤后神经元中表达量随着时间推移逐渐增加,提示上述相关基因可能促进神经元的凋亡,参与了脊髓继发性损伤。电针通过抑制Caspase-3在大鼠脊髓损伤神经元中表达,抑制神经细胞凋亡,减轻了脊髓继发性损伤,促进神经功能恢复。
Objective:With the model rats of acute SCI,the paper investigate how electro-aupuncture affects the expression of neural cell apoptosis and changes of related genes for caspase-3 after spinal cord injury in Rats.The paper tries to show the mechanism of electro-acupuncture on spinal cord injury.Methods:With improved Allen's method to make rats acute SCI,healthy female Wistar rats were randomly divided into four groups,including electro-acupuncture group,Z-DEVD-FMK group,the model group and sham-operation group.The neurofunctional comeback of acute spinal cord injury was measured by TUNEL methods.Results:The result of Z-DEVD-FMK group shows:The number of the positive cells for caspase-3 in Z-DEVD-FMK group was much less than that in the model group at 3d(P0.05).The result of electro-acupuncture group shows:The number of the positive cells for caspase-3 in electro-acupuncture group was much less than that in the model group at the 14th day(P0.05).Conclusion:First: There are neuronal cell apoptosis after spinal cord injury.It is mainly pathology mechanism of secondary spinal cord injury.Second:The number of the positive cells for Caspase-3 is gradually increased with the passage of time.It shows that the above related genes may promote apoptosis of neuron and join the mechanism of secondary spinal cord injury.Electro-acupuncture inhibits the number of the positive cells for Caspase-3 in neuron after spinal cord injury in rats,inhibits neron cell apoptosis,reduces secondary spinal cord injury and promotes recovery of neurofunction.
