促血小板生成素在小胶质细胞炎症模型中的表达及与NF-κB的关系 被引量:1

Expression of TPO and inflammatory cytokines NF-κB in the inflammation model of microglia induced by lipopolysac charide

摘要 目的探讨脂多糖(lipopolysaccharide,LPS)干预小胶质细胞后细胞内NK-κB及促血小板生成素(thrombopoietin,TPO)的分泌。方法将BV2细胞分为6组:(1)空白12 h组;(2)LPS 0.5μg/ml 12 h组;(3)LPS 1μg/ml12 h组;(4)空白24 h组;(5)LPS 0.5μg/ml 24 h组;(6)LPS 1μg/ml 24 h组。采用ELISA法检测细胞内NF-κB和TPO的表达;实时荧光定量PCR法检测细胞内NF-κB及TPO mRNA的表达水平。结果 NF-κB和TPO的表达和mRNA均较空白组升高,且12 h组高于24 h组(P<0.05)。TPO与NF-κB的表达呈显著的正相关(P<0.01)。结论 LPS干预小胶质细胞后NF-κB和TPO表达升高,参与炎症、凋亡和神经元保护等多种调控机制。 Objective To study the secretion of thrombopoietin(TPO) and inflammatory cytokines in microglia cells with the intervention of lipopolysaccharide(LPS).Method BV2 cells were randomly divided into six groups as followings:(1)blank 12h group;(2)LPS 0.5 μg/ml 12 h group;(3)LPS 1 μg/ml 12 h group;(4)blank 24 h group;(5)LPS 0.5 μg/ml 24 h group;(6)LPS 1 μg/ml 24 h group;The expression of intracellular inflammatory cytokines NF-κB and TPO were detected by ELISA;Real-time quantitative PCR was used to check the expression of NF-κB and TPO mRNA level.Results The expression and their mRNA level of NF-κB and TPO in groups with the intervention of LPS were much higher than those in the control group(P〈0.05),moreover,the expression and their mRNA level of NF-κB and TPO in LPS 12 h group were higher than those in 24 h group(P〈0.05).There was a significant positive correlation with the expression of NF-κB and TPO(P〈0.01).Conclusion LPS can significantly increase the expression of NF-κB and TPO in BV2 cells,which may participate in the inflammation,apoptosis and neuronal protection in central nervous system.
