摘要
以ODS-C18(5μm,4.6 mm×150 mm)为色谱柱,庚烷磺酸钠离子对试剂(PIC-B7)和甲醇为流动相并采用梯度洗脱,在流速为1 mL.min-1、柱温为30℃条件下,用波长为275 nm的紫外检测器进行高效液相色谱法(HPLC)检测和分析白牦牛乳中B族维生素的含量.结果表明:白牦牛乳中维生素B3的含量最高(345.588 6μg.(100 mL)-1),维生素B11的含量最低(4.815 7μg.(100 mL)-1);B族维生素的平均回收率为82.98%~97.96%.在本研究所建立的样品前处理方法和HPLC分析色谱条件下,能快速、准确地同时测定白牦牛乳中各种B族维生素的含量,具有重现性好和回收率高的特点.
The high performance liquid chromatography method for a simultaneous contents determination of B vitamins in white yak milk had been developed.Separation and contents determination of B vitamins were achieved by using ODS-C18 column(5 μm,4.6 mm×150 mm),gradient elution with sodium heptanesulfonate(PIC-B7 reagent) and methanol as mobile phase at the flow of 1 mL·min-1,column temperature 30 ℃ and 275 nm UV.The results showed that the maximal and minimal contents of B vitamins were B3(345.588 6 μg·(100 mL)-1) and B11(4.815 7 μg·(100 mL)-1) respectively.The average recover rate of B vitamins was 82.98 %~97.96 %.With the ascertained samples pre-dispose method and chromatography conditions in the study,B vitamins in white yak milk have been simultaneous determined quickly and accurately,and with the advantages of better repeatability and higher recovery.
