摘要
目的评价间歇性高糖对胰岛β细胞的损害及艾塞那肽(Exenatide)的保护作用。方法取大鼠胰岛INS-1细胞分为5组;正常糖(5.5mmol/L)对照组、恒定性高糖(30mmol/L)组和恒定性高糖+Exenatide(100nmol/L)组按分组所述对细胞进行培养,间歇性高糖组和间歇性高糖+Exenatide组采用高糖与正常糖交替(每24h更替1次)培养细胞。培养7d后检测各组细胞内活性氧自由基(ROS)和黄嘌呤氧化酶(XOD)水平,同时检测细胞增殖和凋亡情况。结果与正常糖对照组比较,恒定性高糖组及间歇性高糖组细胞内ROS、XOD水平和细胞凋亡率明显增加(P<0.01),细胞增殖活性明显降低(P<0.01),尤以间歇性高糖组更为明显。恒定性高糖+Exenatide组的ROS、XOD水平和细胞凋亡率明显低于恒定性高糖组(P<0.05),而间歇性高糖+Exenatide组明显低于间歇性高糖组(P<0.01);恒定性高糖+Exenatide组和间歇性高糖+Exenatide组的细胞增殖活性与正常糖对照组无统计学差异。结论间歇性高糖和恒定性高糖均能增加大鼠INS-1细胞内氧化应激水平,从而促进细胞凋亡、抑制细胞增殖,尤以间歇性高糖更为明显;Ex-enatide可显著减轻高糖对细胞的损害。
Objective To evaluate the impairment of islet β cells in intermittent high glucose environment,and the protective effect of exenatide.Methods The rats’ islet β cells were harvested and divided into 5 groups.Cells in the control group were either exposed to normal concentration of glucose(5.5mmol/L),or constant high glucose group to high concentration of glucose(30mmol/L),or in the constant high glucose+exenatide group to high concentration of glucose(30mmol/L) plus exenatide(100nmol/L).The cells in the intermittent high glucose group and intermittent high glucose+exenatide group were alternately cultured in normal or high glucose environment(24 hours for each).The levels of reactive oxygen species(ROS) and xanthine oxidase(XOD) in cells,and the proliferation and apoptosis of cells were determined on day 7 after culturing.Results In comparison to the control group,the levels of ROS and XOD,and the apoptotic rate increased significantly in the constant high glucose group,and especially the intermittent high glucose group(P〈0.01).Lower levels of ROS,XOD and apoptotic rate were found in constant high glucose exenatide group compared with the constant high glucose group(P〈0.05),while the levels of ROS,XOD and apoptotic rate were lower in intermittent high glucose+exenatide group than in the intermittent high glucose group.However,no significant difference in the proliferation was found among the control group,intermittent high glucose+exenatide group and the constant high glucose+exenatide group.Conclusions High concentration of glucose,either in a constant or intermittent manner,may elevate the level of oxidative stress of INS-1 cells,thus leads to an increase in apoptosis and a decrease in proliferation,and it is especially prominant in the intermittent high glucose environment.It is suggested that exenatide may play a protective role on INS-1 cells exposed to high glucose environment.
