摘要
以油茶(Camellia oleifera Abel.)优良无性系的幼胚和子叶为外植体,采用正交试验设计方法,研究了基本培养基、IBA、6-BA和NAA等植物激素共4个因素的不同组合对其幼胚再生体系及子叶胚性愈伤组织诱导的影响,并进一步研究了不同植物激素对体细胞胚再生体系的影响。结果表明,植物激素的选择对幼胚再生体系的建立起着关键作用,NAA促进了幼胚再生苗的形成,IBA的添加反而阻碍了该体系的建立;由数据分析综合评定法,得出幼胚再生植株诱导最适培养基的理论值为WPM+2.0mg/L 6-BA+1.5 mg/L NAA;子叶胚性愈伤组织诱导的最适培养基为WPM+1.0 mg/L 6-BA+0.5 mg/L NAA;子叶胚性愈伤组织的最适分化培养基为MS+2.0mg/L 6-BA+0.01mg/L NAA。
The influence of basic medium,IBA,6-BA,and NAA on the establishment of immature embryos regeneration system and the induction of cotyledon embryo callus was studied by orthogonal design using cotyledon and immature embryos of Camellia oleifera Abel.as the materials.Further influence of different plant growth regulators on regeneration system of cotyledon and young embryo was conducted.The results showed that,the choice of growth hormone played a key role in the establishment of immature embryos regeneration system.NAA boosted the formation of regenerated shoots,but IBA hindered the establishment of the system.According to the analyses,the best medium was WPM+2.0 mg/L 6-BA+1.5 mg/L NAA for the culture of immature young embryos,WPM+1.0 mg/L 6-BA+0.5 mg/L NAA for the culture of cotyledon embryo callus and MS+2.0 mg/L 6-BA+0.01 mg/L NAA for the differentiation of the cotyledon embryo callus.
