摘要
通过测定京尼平交联胶原后的颜色、交联度、吸液率、降解率及细胞毒性的变化,研究交联pH值对京尼平交联胶原制备生物材料的影响.分别于pH5.0,5.5,6.0,6.5,7.0条件下,在0.5%京尼平水溶液中交联24 h,以未交联的胶原作为对照,评价所得材料的颜色、交联度、吸液率、降解率、细胞毒性及组织相容性特点.当pH为5.5时,交联后材料的颜色较浅,且较好的生物相容性以及良好的抗降解性.
The changes of color, crosslinking index, swelling ration, degradation rate, cytotoxicity and biocompatibility of genipin crosslinked collagen were investigated to study the effaction of the different crosslinking pH. The cross-linking reaction parameters of genipin with collagen were all evaluated. The swelling ratio, the degree of cross-linking, and the rate of degradation of cross-linked collagens were measured by use of using ninhydrin assay. The results indicat that genipin is a good cross-linker for the collagen, and pH play an important crucial significant role in the reaction. When the pH of cross-linking reaction is 5.5, the cross-linked collagen would be nearly almost colorless. Additionally, the characters properties of the cross-linked collagen are better compatible, well stability, both have been confirmed by in vitro culture.
