摘要
目的:探讨Lasp1基因与蛋白质水平在口腔鳞状细胞癌(OSCC)组织标本中的表达及其临床意义。方法:分别采用荧光实时定量RT-PCR和免疫组织化学方法,检测Lasp1在30例口腔鳞癌患者的癌组织及癌旁组织标本中mRNA和蛋白质表达水平,其表达水平与临床病理指标之间的关系采用SPSS10.0软件包进行统计分析。结果:口腔鳞癌组织标本中Lasp1 mRNA和蛋白质表达水平均显著高于癌旁组织(P<0.01)。Lasp1蛋白在临床晚期、淋巴结转移阳性病例中表达明显升高,与烟酒嗜好、肿瘤大小及肿瘤分化程度无关。结论:Lasp1在口腔鳞状细胞癌中表达上调,可能与肿瘤的发生、发展和淋巴结转移有关,可以作为一个潜在的生物标志物和治疗的靶标。
Objective: To investigate the gene transcript level and protein expression of Lasp1 in oral squamous cell carcinoma tissues as well as their clinical significance.Methods: The mRNA level and protein expression of Lasp1 in 30 cases of oral squamous cell carcinoma tissues and paracancerous tissues were detected by fluorescent quantitive real-time RT-PCR and immunohistochemistry techniques respectively.The relationship between the Lasp1 expression level and the clinicopathological parameters was analyzed by SPSS10.0 software package.Results: The Lasp1 mRNA transcript level and protein expression in cancerous tissues increased significantly compared to the paracancerous tissues.Though no significant correlation was found between Lasp1 expression and smoking,drinking,T stage and pathological differential degree,OSCC patients with more advanced clinical stages and lymph node metastasis were prone to have higher expression of Lasp1.Conclusion: The results suggest that increased expression of Lasp1 contributes to cancerous progression and lymph node metastasis,and Lasp1 may be potentially used as a biomarker and a therapeutic target for OSCC.
