白术多糖预处理对大鼠肝脏缺血再灌注损伤后肝细胞线粒体结构的影响 被引量:15

摘要 目的探讨白术多糖预处理对大鼠肝脏缺血再灌注损伤后肝细胞线粒体结构的影响。方法将54只成年雄性SD大鼠随机分成3组:假手术组(Sham组)、缺血再灌注组(HIRI组)、白术多糖预处理缺血再灌注组(AMP组),分别于缺血60 min再灌注1 h,6 h,24 h后取材,透射电子显微镜下观察肝细胞的超微结构变化,定量分析线粒体的直径、周长及面积的变化情况。结果电镜下见缺血再灌注组大鼠肝细胞线粒体细胞核皱缩变形,染色质粗糙,核仁浓缩甚至裂解,部分膜破裂,线粒体嵴疏松溶解等,而白术多糖预处理组明显好转。定量分析线粒体的直径、周长及面积,AMP组与HIRI组差异具有显著性(P<0.05),AMP组线粒体损伤程度较HIRI组减轻。结论白术多糖对肝脏缺血再灌注损伤造成的肝细胞线粒体损伤具有显著的保护作用。 Objective To investigate the effect of atractylodes macrocephalaon polysaccharide pre-disposal treatment after ischemia reperfusion injury of liver on the chondriosome structure of hepatic cell in rats.Methods Fifty-four male SD rats were randomly divided into 3 groups: Sham group,ischemia-reperfusion group(HIRI group) and pre-disposal treatment of atractylodes macrocephalaon polysaccharide group(AMP group),the rats were killed at 1,6 and 24 h after 60 min ischemia-reperfusion,the changes of ultramicro-structure of hepatic cell were investigated,then the changes of diameters,perimeters and areas of chondrosomes were quantitatively analyzed.Results Cellular nucleus shrinkage deformity,chromatin asperity,nucleole concentration even spallation,rupture of membrane,crista mitochondriales raritas and dissolve were seen in HIRI group under transmission electron microscope,the hepatic cell of AMP group were improved obviously;the diameters,perimeters and areas of chondrosomes were quantitatively analyzed in AMP group,they were much smaller compared with HIRI group(P0.05),the degree of injury was lightened.Conclusion Atractylodes macrocephalaon polysaccharide has an obviously protection of hepatic cell chondriosome from injury caused by hepatic ischemia reperfusion injury.
