摘要
目的:建立应用紫外光谱快速测定金银花提取物中5种有机酸含量的方法。方法:以HPLC法为参照,测定金银花提取物中绿原酸、咖啡酸、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸的含量。在220~400 nm范围内扫描金银花提取物溶液的紫外吸收光谱,采用偏最小二乘法分别建立样品紫外吸收光谱与5种有机酸含量之间的校正模型,并对模型的预测能力进行考察。结果:5个校正模型对预测集样本的预测值与对照值的相关系数都在0.90以上,相对预测误差分别为1.46%,-2.13%,0.42%,6.40%,3.83%,预测结果准确。结论:本方法操作简便、快速准确,可作为金银花提取物中有机酸含量测定的一种快速分析方法。
Objective: A UV method was established for the rapid determination of 5 kinds of organic acids in Flos Lonicerae extracts.Methods:The HPLC method was used as the reference method to determine the contents of chlorogenic acid,caffeic acid,3,4-dicaffeoylquinic acid,3,5-dicaffeoylquinic acid and 4,5-dicaffeoylquinic acid in the Flos Lonicerae extracts.UV absorption spectrum was scanned at the 220-400 nm wavelength range.The PLS models was developed to correlate the UV spectra with the corresponding values determined by the reference method.Then the performance of calibration models was validated.Result:The correlation coefficients of the validation set were all above 0.90,and the relative predicted errors for the 5 kinds of organic acid were 1.46%,-2.13%,0.42%,6.40% and 3.83%,respectively.Conclusion: Owing to its simple,fast and accurate properties,the presented method could be a method for the rapid determination of organic acids in Flos Lonicerae extracts.
