摘要
目的建立HPLC-DAD法同时测定小儿清解颗粒中9种成分(新绿原酸、绿原酸、隐绿原酸、獐芽菜苷、断氧化马钱素、连翘酯苷A、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸)的含量。方法采用Agilent Zorbax SB C18色谱柱(250 mm×4.6 mm,5μm),流动相为0.1%甲酸水溶液-乙腈,梯度洗脱,流速为1.0 mL·min^-1,新绿原酸、绿原酸、隐绿原酸、连翘酯苷A、3,4-二咖啡酰奎宁酸、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸的检测波长为325 nm,獐芽菜苷、断氧化马钱素为240 nm,柱温30℃。结果 68 min分析时间内9个成分分离度良好,在各自的检测范围内浓度与峰面积呈良好的线性关系,r均〉0.9995,加样回收率为95.7%-100.6%,RSD均≤2.1%。应用所建立的方法同时测定了2个厂家生产的小儿清解颗粒中各成分的含量,其结果有一定差异。结论该法简便可行,结果可靠,可为本制剂多指标成分的质量控制提供参考方法。
Objective To establish an HPLC-DAD method for the simultaneous determination of 9 components(neochlorogenic acid, chlorogenic acid, cryptchlorogenic acid, sweroside, secoxyloganin, forsythoside A, 3, 4-dicaffeoylquinic acid, 3, 5-dicaffeoylquinic acid and 4, 5-dicaffeoylquinic acid) in Xiaoer Qingjie granules. MethodsSamples were performed on an Agilent Zorbax SB C18 analytical column(250 mm×4.6 mm, 5 μm) with a gradient elution of 0.1% formic acid and acetonitrile at a flow rate of 1.0 mL · min^- 1. The DAD detector wavelength was 325 nm for neochlorogenic acid, chlorogenic acid, cryptchlorogenic acid, forsythoside A, 3, 4-dicaffeoylquinic acid, 3, 5-dicaffeoylquinic acid and 4, 5-dicaffeoylquinic acid, and 240 nm for sweroside and secoxyloganin. The column temperature was 30 ℃. Results The 9 components were well separated with good linearity between the concentration of each component and peak area within 68 min. The value of r was higher than 0.9995 for all analytes. The recoveries were 95.7%- 100.6%, and RSDs were less than 2.1%. The method was successful in simultaneously determining 9 components in Xiaoer Qingjie granules from 2 manufacturers with great variations in the determination results.Conclusion The method is simple and reliable which can provide reference for the quality control of multi-index components in Xiaoer Qingjie granules.
