摘要
目的建立HPLC-MS/MS法测定三黄片中盐酸小檗碱。方法采用Agilent C18(250mm×4.6mm,5μm)色谱柱;以乙腈-10mM乙酸铵0.1%甲酸溶液(80:20)为流动相;流速1.0mL.min-1;质谱检测器:MRM模式(336.1-320.1);柱温为25℃。结果盐酸小檗碱在800~1ng.mL-1时呈良好线性关系(r=1.000)。盐酸小檗碱平均加样回收率为100.1%,RSD为1.3%。结论该方法简单、快速、准确,可用于三黄片中盐酸小檗碱含量测定。
OBJECTIVE To develop HPLC-MS/MS method for determination of berberine hydrochloride in San-Huang tablets.METHODS The column was Agilent C18(250mm×4.6 mm,5μm);the mobile phase was acetonitrile-a buffer solution consisting of 0.1% acetic acid and 10 mmol·L-1 ammonium acetate(80∶20);the flow rate was 1.0mL·min-1;MS detector:MRM mode((336.1-320.1);the column temperature was 25℃.RESULTS The linear range of berberine hydrochloride was 800~1ng·mL-1(r=1.000),the average recovery was 100.1% and relative standard deviation was 1.3%.CONCLUSION the method is simple,rapid,accurate,and can be applied to the determination of berberine hydrochloride in San-Huang tablets.
