转录因子T—bet在胃癌组织中的表达及其与预后的关系 被引量:5

Expression of transcription factor T-bet in the gastric carcinoma tissues and its correlation with prognosis

摘要 目的 探讨T-box基因家族的新型转录因子T-bet在胃癌组织中的表达及其与预后的关系.方法 应用免疫组织化学染色技术检测201例胃癌患者手术标本中T-bet阳性淋巴细胞的浸润数量,采用SPSS 13.0统计软件分析.结果 胃癌组织中T-bet阳性淋巴细胞的浸润数量为(15.2±9.5)个/高倍视野(HPF);单因素分析显示,T-bet阳性淋巴细胞的浸润数量与胃癌患者的性别、年龄、肿瘤的发生部位、组织类型、复发、病理分级、淋巴结转移无明显相关;但肿瘤大小、浸润深度、TNM分期与T-bet阳性淋巴细胞的浸润数量差异有统计学意义(P<0.05).肿瘤直径<5 cm组T-bet阳性淋巴细胞的浸润数量(16.9±11.8)个/HPF高于肿瘤直径≥5 cm组(13.0±6.7)个/HPF,差异有统计学意义(t=2.35,P<0.05).未侵入肌层组T-bet阳性淋巴细胞的浸润数量(20.2±14.6)个/HPF明显高于侵入肌层组(14.1±8.3)个/HPF,差异有统计学意义(t=2.50,P<0.05).随着肿瘤TNM分期的增加,T-bet阳性淋巴细胞的浸润数量有明显逐步减小的趋势,差异有统计学意义(F=3.40,P<0.05).Kaplan-Meier生存分析结果显示,T-bet高度浸润组(χ≥19.15)总生存期优于低度浸润组(χ<19.15)的趋势,差异无统计学意义(χ2=1.90,P>0.05).T-bet高度浸润组的中位生存时间比低度浸润组长,两组患者的中位生存时间分别为55.0个月(95%CI:28.2~81.8)和37.0个月(95%CI:29.6-44.4).结论 T-bet阳性淋巴细胞的浸润数量与胃癌的预后相关. Objective To analyze the expression of T-bet as a new transcription factor of T-box gene family in the gastric carcinoma tissues and its correlation with prognosis.Methods The number of T-bet positive lymphocytes in the surgically resected specimens of gastric cancer tissues was detected by using immunohistochemistry in 201 cases of gastric carcinoma,and data were analyzed by using SPSS 13.0.Results The number of T-bet positive lymphocytes in gastric carcinoma tissues was ( 15.2 ± 9.5 )/high powered field.Single-factor analysis demonstrated that the number of T-bet positive lymphocytes was not related with sex,age,tumor site,histological types,recurrence,pathological grades or lymph node metastasis,but with the tumor size,invasion depth or TNM staging.The number of T-bet positive lympbocytes was greated in the tumor diameter 〈5 cm group [( 16.9 ± 11.8)/high powered field]than in the tumor diameter ≥5 cm group [(13.0 ± 6.7 )/high powered field],with the difference being statistically significant (t =2.35 ,P 〈0.05 ).The number of T-bet positive lymphocytes was obviously higher in the non-invasive group [( 20.2 ± 14.6 )/higher powered field]than in the invasive group [(14.1 ± 8.3 )/high powered field],with the difference being statistically significant ( t = 2.50,P〈0.05 ).With the increase of TNM staging,the number of T-bet positive lymphocytes has a gradually descendent trend ( F = 3.40 ,P 〈0.05 ).Kaplan-Meier analysis revealed that overall survival of the patients was longer in more infiltrative group(χ≥19.15) than in lesser infiltrative group (χ〈19.15) (χ2 = 1.90,P〉0.05).The median survival time in more infiltrative group (55.0 months) (95% CI:28.2-81.8) was significantly longer than in lesser infiltrative group ( 37.0 months) ( 95% CI:29.6-44.4 ).Conclusion There was a correlation between the number of T-bet positive lymphocytes and the prognosis of gastric carcinoma.
