塞来昔布联合羟基喜树碱对胃癌细胞的生长抑制作用及机制探讨 被引量:1

摘要 目的观察塞来昔布联合羟基喜树碱对胃癌细胞SGC-7901的抑制作用。方法用四甲基偶氮唑蓝(MTT)法观察两药单用及序贯用药对胃癌细胞的抑制作用,用流式细胞仪检测塞来昔布对胃癌细胞诱导凋亡作用。结果 5μg/L羟基喜树碱与25μmol/L塞米昔布联合使用时表现为联合用药组对胃癌细胞的抑制作用明显强于两单药组,5μg/L羟基喜树碱与12.5μmol/L塞来昔布联合时,表现为不同的用药顺序,其抑制作用有显著性差异。流式细胞仪检测发现塞来昔布对胃癌细胞有诱导凋亡作用。结论塞来昔布通过诱导细胞凋亡对胃癌细胞SGC-7901有生长抑制作用。两药联合可能增强对胃癌细胞的抑制作用。 Objective To investigate the growth-inhibiting effect of celecoxib combined with hydroxycamptothecin(HCPT) on human gastric cancer cell line SGC-7901.Methods The cells were treated with 12.5-25 μmol/L of celecoxib combined with HCPT for 48 hours.MTT assay was used to observe the inhibitive effects on the gastric cancer cells when the two drugs were administered in combination and in sequence.Flow cytometry was used to detect cell apoptosis following celecoxib treatment.Results Celecoxib could inhibit the proliferation of gastric cells in vitro.The combined administration of celecoxib(12.5 μmol/L) with HCPT(5 μg/L) had significantly more potent inhibitive effects than those groups with individual administration of either of the drugs.There were significant differences between the combination groups with different sequences of HCPT and celecoxib administration.Flow cytometry revealed that celecoxib could induce apoptosis in the gastric cancer cells.Conclusion Celecoxib has inhibitive effects on human gastric cancer cell line SGC-7901 by inducing apoptosis of the gastric cells.The celecoxib-HCPT combination therapy could effectively enhance the inhibitive effects on gastric cancer cells.
