摘要
目的探讨PARP-1抑制剂3-aminobenzamide(3-AB)对骨肉瘤U2OS细胞增殖及凋亡的影响。方法常规培养骨肉瘤U2OS细胞,采用CCK-8法检测U2OS细胞的增殖和流式细胞术检测细胞凋亡,并用caspase-3活性测定试剂盒检测caspase-3活性和Western-blot技术检测Bcl-2、Bax的表达。结果 3-AB能抑制骨肉瘤细胞的增殖,且与剂量和时间呈相关性,流式细胞术检测结果显示3-AB能促进骨肉瘤细胞的凋亡,且呈时间相关性,3-AB刺激早期caspase-3活性增高,Western-blot显示3-AB刺激后随时间延长Bax的表达量逐渐升高,而Bcl-2的表达量则轻度升高后降低。结论 PARP-1抑制剂3-AB能抑制骨肉瘤细胞的增殖,促进其凋亡,这为PARP-1作为骨肉瘤治疗的新靶点提供了初步实验依据。
Objective To investigate the influence of PARP-1 inhibitor(3-aminobenzamide,3-AB) on the proliferation and apoptosis of osteosarcoma cells.Methods The U2OS cells were routinely cultured.CCK-8 was used to analyze cell proliferation,apoptosis was assessed by flow cytometry,the caspase-3 activity was measured by caspase-3 colorimetric assay kit,and the expression of Bcl-2 and Bax was detected by using Western blot.Results 3-AB suppressed the proliferation of osteosarcoma cells in a dose-and time-dependent manner.It also increased apoptosis ratio of osteosarcoma cells,after treated with 3-AB,the caspase-3 activity in osteosarcoma cells was elevated,the expression of Bax increased,and the expression of Bcl-2 first slightly increased and then decreased.Conclusion 3-AB suppresses cell growth and induces apoptosis in human osteosarcoma cells.Our study provided a preliminary evidence by using PARP-1 as a new target for osteosarcoma treatment.
