摘要
目的:研究转化生长因子-β1(TG9F-β1)、白细胞介素-2(IL-2)、白细胞介素-4(IL-4)在溴隐亭致流产大鼠黄体及血清的表达水平,探讨流产的发生机制。方法:将孕雌性wistar大鼠随机分为3组,正常孕组、注射溴隐亭组、注射溴隐亭+γ-干扰素(IFN-γ)抗体组,3组均于孕12天时,腹主动脉采血后心内灌注固定,酶联免疫吸附试验(ELISA)测定各组血清TGF-β1、IL-2、IL-4水平,免疫组化SP法检测TGF-β1、IL-2、IL-4在各组大鼠卵巢黄体的表达并进行图像分析,分别对上述结果进行单因素方差分析及t检验。结果:血清与卵巢黄体阳性细胞OD值的变化趋势基本一致,注射溴隐亭组,IL-2水平较正常孕组升高(P<0.05)、TGF-β1、IL-4水平较正常孕组降低(P<0.05);注射溴隐亭+IFN-γ抗体组,血清IL-2水平较注射溴隐亭组有所降低(P<0.05),但仍高于正常孕组(P<0.05),而TGF-β1、IL-4水平有所升高(P<0.05),基本达正常水平。结论:给孕鼠注射溴隐亭及IFN-γ抗体后,TGF-β1、IL-2、IL-4在血清及卵巢黄体的表达有一定改变,此改变可能与流产的发生有一定的关系。
Objective:To investigate the expression of transforming growth factor β1 (TGF-β1), IL-2 and IL-4 in luteum body and serum in induced abortion rats by bromocriptine,and to explore the mechanism of abortion. Methods:Pregnant wistar rats were randomly divided into three groups: normal pregnancy group, bromocriptine injection group,bromocriptine and I FN-y antibody injection group. On the 12th day after pregnancy,the blood was collected from abdominal aorta followed by intracardiovascular perfusion fixation,serum level were checked by ELISA,while TGF-β1 ,IL-2 and IL-4 in luteum body by SP immunochemistry. Statistical analysis was done by univariate analysis and t-test. Results :The changes of OD value in corpora luteum positive cells and serum level showed similar tendency. For the bromocriptine injection group, IL-2 level increased markedly than that in normal pregnancy( P 〈0.05), while transforming growth factor β1, IL-4 level decreased significantly (P 〈 0.05). For bromocriptine and I FN-y antibody injection group, IL-2 level in serum decreased compared with that in bromocriptine group ( P 〈 0.05), but still higher than that in normal pregnancy group ( P 〈0.05) ;while TGF-β1 and IL-4 level increased back to normal levle ( P 〈 0.05). Conclusions:After injection of bromocriptine and IFN-y, antibody,the expressions of TGF-β1, IL-2 and IL-4 in serum and corpora luteum have some changes, and these changes probably associate with genesis of abortion.
