Effect of transplantation of neural stem cells on connexin43 protein expression in cortex of SD rat with brain weight-dropping injury

摘要 目的观察神经干细胞(neural stem cells,NSCs)移植治疗大鼠脑损伤前后大脑皮质神经元的缝隙连接蛋白43(Connexin 43,Cx43)表达变化,探讨NSCs移植治疗对脑外伤功能恢复的作用。方法体外培养NSCs,自由落体撞击大鼠左脑大脑皮质运动感觉区以制作脑外伤模型,采用免疫组织化学、RT-PCR来检测脑外伤后2w和4w及脑外伤神经干细胞移植后2w和4w时的Cx43的表达变化。结果大鼠大脑皮质Cx43的表达水平创伤组明显多于sham组,移植组Cx43的表达水平明显多于创伤组(P<0.05)。结论 Cx43的表达变化可能与NSCs移植对脑外伤恢复作用有关。 Objective To study the effect of transplantation of neural stem cells on connexin43(Cx43) protein expression in cortex of SD rat with brain weight-dropping injury. Methods Neural stem cells(NSCs) were cultured in vitro and transplanted into cortical sensoniomotor area of rat injured by weight-dropping within 24 hours. Immunohistochemistry and RT-PCR were used to detect the expression of Cx43. Results The expression level of Cx43 in the cortical area was increased in injuried group than in sham, the expression of Cx43 in the cortical area was increased after transplantation of NSCs( 0.05). Conclusion The expression of Cx43 might be related to the repair of NSCs on the brain injury.
