摘要
为了得到纯净的、具有较高生理和药理活性的单体人参皂苷Rh1,从三七中提取总皂苷,并对三七总皂苷进行分离纯化获得人参皂苷Rh1单体。分别经过乙醇浸提、石油醚脱脂、水饱和正丁醇萃取、大孔吸附树脂脱糖脱色及硅胶层析,从三七总皂苷中分离得到人参皂苷Rh1。结果表明,1 000 g三七干燥根经醇提、脱脂、萃取和脱糖脱色可制得三七总皂苷76.2 g,提取率为7.62%。25 g三七总皂苷以氯仿-甲醇溶液[V(氯仿)∶V(甲醇)=8.5∶1.5]作为流动相洗脱,经硅胶层析得人参皂苷Rh1单体0.36 g,得率为1.44%,高效液相色谱检测其纯度为66.08%。
The ginsenoside Rh1 in Panax notoginseng was isolated by silica gel chromatography.76.2 g total notoginsenoside was obtained from 1 kg Panax notoginseng,the yield of total notoginsenoside was about 7.62%.0.36 g ginsenoside Rh1 was separated from 25 g total notoginsenoside by silica gel chromatography using chloroform and methanol at 8.5∶1.5 as the mobile phase,the yield of ginsenoside Rh1 was 1.44%,the purity was 66.08% detecting by HPLC.
