摘要
目的:了解ERK-MAPK信号通路在颅骨缝牵张成骨中的活化规律。方法:建立大鼠颅骨矢状缝牵张培养模型,运用WesternBlot及免疫组织化学技术,观察p-ERK1/2在骨缝内的表达。结果:在牵张力的作用下,p-ERK1/2表达量于10min达到顶点,随后表达下降并在80min时维持在初始状态的低水平,并且阳性细胞主要集中在骨缝边缘区域。结论:ERK-MAPK信号通路的激活与张应力相关,该通路活化区域和牵张过程中细胞增殖、分化高表达区域一致。表明ERK-MAPK信号通路参与了骨缝内细胞力学信号向生物学信号的转化。
Objective:To observe whether the ERK-MAPK signal pathway participates in suture distraction osteogensis. Methods:After the establishment of the organ cultured calvarial suture distraction osteogenesis models,we applied Western blot and immunohistochemisty methods to analysis the expression pattern of ERK-MAPK signal pathway in sutures. Results:Under the distraction,p-ERK1/2 reached the peak in 10min and since then kept in a lower level as the start,and the cells adjacent to the bone edges were mainly p-ERK1/2 positive. Conclusion:The activation of ERK-MAPK signal pathway is related to distraction and may join the process of the conversion from mechanical stress to biology response.
