摘要 目的:检测整合素β1(integrinβ1)在下颌牵张成骨过程中的分布和表达,初步探讨整合素β1在张应力信号传导过程中的作用。方法:对12只大耳白兔行双侧下颌骨骨切开术,并安置口外牵张器,经7d间歇期后,按1mm/d的速率将下颌骨延长6mm。在间歇期末,牵张结束当天及牵张结束后第7天和第14天分别处死3只动物,取牵张区新生组织作免疫组化检测不同时间点整合素β1的分布特征和表达水平,并用染色强度等级分析法对免疫组化表达水平的变化进行评价。结果:整合素β1免疫组化阳性染色主要位于牵张间隙内的间充质细胞和新生骨小梁表面的成骨细胞。间歇期末,多数间充质细胞和成骨细胞免疫组化染色呈弱阳性;牵张结束时,染色强度显著上升;然而,固定到第7天和第14天,染色强度明显弱于牵张结束时。结论:整合素β1可能在牵张成骨时将外界的张应力传递至感应细胞内并刺激成骨细胞系增殖分化的过程中起重要作用。 Objective: To observe the distribution and expression of integrin β1 during mandibular distraction osteogenesis.To investigate the function of integrin β1 in the transduction of mechanical stimuli to sensory cell during mandibular osteodistraction. Methods: Bilateral mandibular osteotomies were performed in 12 rabbits. The mandibles were lengthened 6mm using a custom-made distractor with a rate of 1mm/d. 3 rabbits were killed respectively at the end of delay period and 0、7 and 14 days, distraction. The distracted calluses from all animals were harvested and processed for immunohistochemistry staining for integrin β1. Semi-quantitative analysis was used to evaluate immunohistochemical staining. Results: Immunohistochemical staining of integrin β1 was localized in the mesenchymal cells at distraction gap and osteoblast lining on the surface of trabecular columns. The majority of mesenchymal cells and osteoblastic cells failed to immmunodetectable reactions at the end of latency. After 0 day's distraction, a marked gradient increase of expression was readily visible in the regenerated tissue. After the 7 and 14 days distraction, the intensity of its expression was significantly weaker than that of 0 day. Conclusion: The results suggest that integrin β1 may play an important role in the transduction of mechanical stimuli to sensory cell and interpret intracellular proliferation and differentiation messages during distraction osteogenesis.
