噻环乙胺及小型猪复合麻醉剂(XFM)对大鼠不同脑区神经肽含量的影响 被引量:1

Effects of tiletamine and its compound preparationon(XFM)on neuropeptide contents in different encephalic region of rats

摘要 通过研究噻环乙胺及XFM麻醉下大鼠不同脑区OFQ、dynA和SP含量的变化,探讨噻环乙胺及XFM中枢麻醉作用可能的机理。Wistar大鼠84只,先随机抽取12只为对照组。其余随机均分为噻环乙胺组和XFM组,每组又随机均分为麻醉组、恢复Ⅰ组和恢复Ⅱ组3个亚组。采用酶联免疫吸附试验(ELISA)测定各脑区内OFQ、dy-nA和SP的含量。结果显示,腹腔注射(ip)噻环乙胺2 mL/kg后,麻醉组大鼠大脑皮层和丘脑内OFQ含量显著降低(P<0.05),dynA和SP含量均显著增加(P<0.05或P<0.01);脑干内OFQ含量显著降低(P<0.05),dynA含量显著增加(P<0.05),SP含量无显著变化。恢复Ⅰ组和恢复Ⅱ组大鼠上述各脑区内OFQ、dynA和SP含量均显著恢复(P>0.05);麻醉全过程中,海马和小脑内OFQ、dynA和SP含量均无显著变化。ip XFM 2 mL/kg后,麻醉组,大脑皮层内dynA含量显著增加(P<0.01),OFQ和SP含量均无显著变化;丘脑内OFQ含量显著降低(P<0.01),dynA和SP含量均显著增加(P<0.01);恢复Ⅰ组和恢复Ⅱ组,上述各脑区内OFQ、dynA和SP含量均显著恢复(P>0.05)。结果表明,噻环乙胺和XFM对不同脑区OFQ、dynA和SP含量的影响可能是其产生全麻作用的重要机理之一;噻环乙胺麻醉中枢作用可能与降低大脑皮层、丘脑和脑干内OFQ,增加大脑皮层和丘脑内dynA和SP及脑干内dynA的含量有关;而XFM则可能与降低丘脑内OFQ,增加丘脑内dynA和SP及大脑皮层内dynA的含量有关。 To explain the possible molecular mechanisms of tiletamine and its compound preparationon(XFM) anesthesia on the central nervous system by investigating the changes of OFQ,dynA and SP contents in different encephalic regions in rats.12 Wista rats were allocated randomly from 84 Wista rats as control group.The rest rats were divided randomly into tiletamine and XFM groups,and each group of animals were divided randomly into maintenance of anesthesia group,recovery from anesthesiaⅠgroup and recovery from anesthesiaⅡgroup.The OFQ,dynA and SP contents were measured by enzyme-linked immunosorbent assays(ELISA).After injected intraperitoneally with tiletamine 2 mL/kg,the OFQ contents in the cerebral cortex and the thalamus in the anesthesia group were obviously decreased as compared with control group(P0.05),the dynA and SP contents were obviously raised(P0.05 or P0.01);the OFQ contents in the brain stem in the anesthesia group were obviously decreased as compared with control group(P0.05),the dynA contents were obviously raised(P0.05),the SP contents were not obviously changed.The OFQ,dynA and SP contents in the above encephalic regions in the recovery from anesthesiaⅠgroup and anesthesiaⅡgroup were obviously revived as compared with control group(P0.05);The OFQ,dynA and SP contents in the hippocampus and cerebellum were not obviously changed in the process of general anesthesia of tiletamine.After injected intraperitoneally with XFM 2 mL/kg,the dynA contents in the cerebral cortex in the anesthesia group were obviously raised as compared with control group(P0.01),the OFQ and SP contents were not obviously changed;The OFQ contents in the thalamus in the anesthesia group were obviously decreased as compared with control group(P0.01),the dynA and SP contents were obviously raised(P0.01).The OFQ,dynA and SP contents in the above encephalic regions in the recovery from anesthesiaⅠgroup and anesthesiaⅡgroup were obviously revived as compared with control group(P0.05);
