摘要 目的研究p27kip1及相关分子Jab1蛋白表达与喉鳞状细胞癌生物学行为的关系,探讨它们在喉鳞癌发生、发展中的作用。方法采用免疫组织化学二步法测定50例喉鳞状细胞癌(LSCC)、15例癌旁组织中p27kip1和Jab1的表达情况。结果 p27kip1蛋白在LSCC中低表达(44.0%);Jab1蛋白在LSCC中的表达高于癌旁喉黏膜组织,差异有显著性(P<0.01),并且与增殖指标Ki-67的表达呈正相关(P<0.05)。Jab1蛋白表达与TNM分期、分化程度及淋巴结转移相关(P<0.05)。p27kip1与Jab1表达水平呈负相关(P<0.05)。结论 Jab1作为p27kip1的负性调控分子,可能参与LSCC的发生发展过程,联合检测p27kip1及Jab1的表达有助于综合评估LSCC的生物学行为。 Objective To investigate the expression of p27^kip1 and Jab1, and the correlation between the expression and biological behaviors of laryngeal squamous cell carcinomas (LSCC). MethodsThe expression of p27^kip1 and Jab1 in them were examined by immunohistochemistry using highly specific antibodies on formalin-fixed, paraffin-embedded tissue section from 50 LSCC, 15 para-cancerous tissue. ResultsWe observed the more significantly increased expression of Jab1 in LSCC group than those in group of para-cancerous tissue. A negative correlation had been noted between expression of p27^kip1 and Jab1. Over-expression of Jab1 in LSCC strongly correlated with tumor differentiation, the TNM stage and lymph node metastasis. Conclusion These results suggested that Jab1, as a negative regulator of p27^kip1, may have an important role in the development of aggressive behavior in LSCC. Combined detection of the expression of p27^kip1 and Jab1 may be useful in evaluating the biological behaviors of LSCC comprehensively.
