摘要
改良菘蓝现有品种,丰富物种资源,增加其药用成分的含量。采用正交试验法考察无菌水浸泡时间,秋水仙素浓度和浸泡时间对菘蓝种子多倍体诱导率的影响;用L9(33)正交试验筛选出诱导菘蓝多倍体植株的最佳条件。在考察的因素中,对菘蓝多倍体植株诱导率的影响程度为秋水仙素的浓度>秋水仙素浸泡时间>无菌水浸泡时间。多倍体诱导的最佳条件为A2B2C3,即种子经无菌水浸泡24 h,再用0.2%秋水仙素处理36 h后诱变率可达73.91%;经植物形态组织学和染色体数检测,诱导后的植物材料具有明显的多倍体特征。
This experiment aimed at improving the existence breed of Isatis indigotica Fort. , enriching its inheritencable breed resource, in- creasing its medicinal composing. The induction rate of Isatis indigotica Fort. optimized condition( The time being soaked in water,consistency of colchcine and the time being soaked in colchcine) was determined by Orthogonal Experiment. A best combination was sifted through the crossed test of L9 (3^3 ). The order of factors to affect the induction rate was consistency of colcheine 〉 the time being soaked in eolehcine 〉 the time being soaked in water. The optimum combination were A2B2C3 , of which rate could reach 73.91% after treatment with water in 24 h and then 0.2 % colehcine in 36 h. The morphology and chromosomal number proved that the polyploid induetion was effective.
